The Englishman, who claimed his first PGA Tour title last August, carded five birdies against a single bogey to reach eight under for the tournament.
At the top of the leaderboard sits South Korea's Sungjae Im, who posted a steady two-under 69 to hold a slim one-shot advantage at nine under. Fleetwood is in hot pursuit, with compatriot Alex Fitzpatrick and American Justin Thomas lurking just two shots back at seven under.
The Fitzpatrick family storyline adds extra intrigue, as Alex finds himself nine strokes clear of his older brother Matt, the world number three who is struggling at two over par.
Masters champion Rory McIlroy is also firmly in the mix after a bogey-birdie rollercoaster. The Northern Irishman bounced back from dropped shots at the second and 18th holes with six birdies en route to a 67, putting him at five under alongside American Rickie Fowler. Fowler made the biggest move of the day, firing a blistering 63 with nine birdies to vault up the leaderboard.
With no cut at this event, all players will compete through the weekend at the challenging Quail Hollow layout. This tournament serves as the final tune-up before the US PGA Championship, which gets underway at Aronimink Golf Club in Pennsylvania from May 14-17.
